Azaspiracids (AZAs) are lipophilic biotoxins produced by marine algae that can contaminate shellfish and cause human illness. The European Union (EU) regulates the level of AZAs in shellfish destined for the commercial market, with liquid chromatography-mass spectrometry (LC-MS) being used as the official reference method for regulatory analysis. Certified reference materials (CRMs) are essential tools for the development, validation, and quality control of LC-MS methods. Ostreopsis cf. ovata is a toxic marine benthic dinoflagellate responsible for harmful blooms affecting ecosystem and human health, mostly in the Mediterranean Sea. In this study we report the occurrence of a summer O. cf. ovata bloom in Currais, a coastal archipelago located on the subtropical Brazilian coast (~25° S). This bloom was very similar to Mediterranean episodes in many aspects: (a) field-sampled and cultivated O. cf. ovata cells aligned phylogenetically (ITS and LSU regions) along with Mediterranean strains; (b) the bloom occurred at increasing temperature and irradiance, and decreasing wind speed; (c) cell densities reached up to 8.0 × 104 cell cm−2 on fiberglass screen and 5.6 × 105 cell g−1 fresh weight on seaweeds; (d) and toxin profiles... |

ABSTRACT From September 2014 to November 2015, 30 Paralichthys orbignyanus specimens were obtained from Sepetiba Bay in the municipality of Rio de Janeiro, State of Rio de Janeiro, Brazil. The fishes were necropsied and cestode larvae were recovered from the abdominal cavity, mesentery, and serosas of the stomach, intestine, spleen, liver, kidney and gonads. Nineteen fish were parasitized by a total of 90 trypanorhynch plerocerci. Based on morphological characters, these were identified as Pterobothrium crassicolle. The parasitism indices for P. crassicolle on P. orbignyanus, were 63.3% for prevalence, 4.7 for mean intensity, 3 for mean abundance and 1 to 18 for range of infection. The neuroblastoma cell-based assay (CBA-N2a) is widely used for the detection of marine biotoxins in seafood products, yet a consensus protocol is still lacking. In this study, six key parameters of CBA-N2a were revisited: cell seeding densities, cell layer viability after 26 h growth, MTT incubation time, Ouabain and Veratridine treatment and solvent and matrix effects. A step-by-step protocol was defined identifying five viability controls for the validation of CBA-N2a results.
